They likely would have known about it (probably few others) and it probably did effect their relationship. For the worse. They're the epitome of what each other hate... being related to each other just drummed that anomosity in all the more. Almost like the taint or humiliation is carried in the blood. Not unlike Sirius felt about his family or Mrs. Black felt about him, Andromeda or the Weasleys.

Not necessarily. If both the Potters and the Snapes tried to bury the relation, no one may even know about it these days but a select few. I could see Dumbledore being in that small clique. DD's relationship with Snape tends to be strained at best most of the time and this information doesn't really benefit Harry either (but would probably needlessly irritate / upset him if he learned of it), thus I could understand why - if this were true - he hasn't told Harry about it.

James not acknowledging his relation to Snape, vice versa, would also be a nice parallel to Harry not admitting he was nearly sorted into Slytherin. I don't think he's even told Ron or Hermione about that yet. James may not have told Sirius or his other friends about being related to Snape either.
